چکیده

By means of ultrasonication two fractions of the carboxymethylated chitin–glucan complex isolated from the cell walls of 13 the filamentous fungus Aspergillus niger were obtained. Elemental analysis as well as C NMR investigation of the high-molecular weight (M 680 kDa) and low-molecular weight (M 75 kDa) fractions revealed their essentially different w w chitin content. HPLC analysis of the fractions produced by ultrasonic treatment of different duration, combined with nitrogen assay showed steadily increasing chitin content in the faster eluted fraction that allows one to suggest the different susceptibility of the two components of the chitin–glucan complex to the ultrasonication.  1999 Elsevier Science B.V. Chemical morphology of glucan and chitin in the cell wall of the yeast phase of Paracoccidioides brasiliensis.

Short and thick fibers were observed on the outer surface of the yeast phase of Paracoccidioides brasiliensis, and long and thin fibers were seen on the inner surface. The long fibers disappear with chitinase treatment and are composed of chitin. The short fibers disappear under alkali treatment and are composed of alpha-glucan. Comparisons with alpha-(1 --> 3)-glucan isolated from Aspergillus ...

Sonication of chitin-glucan, preparation of water-soluble fractions and characterization by HPLC.

A water-insoluble chitin-glucan complex, isolated from the mycelium of Aspergillus niger, was swollen in various aqueous media and treated subsequently by high-energy sonication. The concentration of the resulting water-soluble polysaccharide fractions was dependent on the swelling medium, the amount of the chitin-glucan complex in the suspension, and on the time of sonication. The yields of wa...

Physicochemical properties and bioactivity of fungal chitin and chitosan.

Chitinous material was extracted from mycelia of Aspergillus niger and Mucor rouxii grown in yeast peptone dextrose broth for 15 and 21 days, respectively. The extracted material was characterized for purity, degree of acetylation, and crystallinity and tested for antibacterial and eliciting properties. The maximum glucosamine level determined in the mycelium of A. niger was 11.10% dw and in th...

Production and Characterization of Fungal Chitin and Chitosan

Chitin is a biopolymer. Theoretically, it is a homopolymer of acetyl-glucosamine. Chitosan is the deacetylated product of chitin. Chitin and chitosan have many applications in water purification, agriculture, food and pharmaceutical industries. Currently, industrial production for chitin and chitosan is from the shell wastes. The objective of this study was to evaluate the feasibility of chitin...

Reshuffling of Aspergillus fumigatus cell wall components chitin and β-glucan under the influence of caspofungin or nikkomycin Z alone or in combination.

Chitin and β-glucan are major cell wall components of Aspergillus spp. We investigated the antifungal activity of chitin synthesis inhibitors nikkomycin Z, polyoxin D, flufenoxuron, lufenuron, and teflubenzuron, alone and combined with the β-glucan synthesis inhibitor caspofungin. Only nikkomycin Z and caspofungin were found to act synergistically. The nikkomycin Z-induced chitin decrease corre...
